Mucopolysaccharide polysulfate promotes microvascular stabilization and barrier integrity of dermal microvascular endothelial cells via activation of the angiopoietin-1/Tie2 pathway

BackgroundMucopolysaccharide polysulfate (MPS) is a heparinoid and MPS-containing formulations are widely used as moisturizers for dry skin to treat peripheral vascular insufficiency. Although MPS has therapeutic effects in diseases with microvascular abnormalities, the of on function remain incompletely understood.ObjectiveThe aim this study was evaluate functional activities human pericytes (HPC) dermal endothelial cells (HDMEC) vitro, permeability skin.MethodsThe protein expression angiopoietin (Ang)-1 HPC, platelet-derived growth factor-BB (PDGF-BB) phosphorylated tyrosine-protein kinase receptor 2 (Tie2) HDMEC were measured presence or absence MPS. The barrier evaluated by expressions claudin-5 (VE)-cadherin, transendothelial electrical resistance (TEER).ResultsIn dose-dependently enhanced Ang-1 secretion, which activated Tie2 HDMEC. In HDMEC, significantly increased production PDGF-BB, important recruitment HPC endothelium, phosphorylation Tie2, results activation Ang-1/Tie2 signaling . tight junction TEER Moreover, intradermal injection prevented factor-induced increase mouse skin.ConclusionWe found that promoted stabilization integrity via activation. These suggest might improve abnormalities various accompanied disturbances signaling.
